Phylogenetic analysis of Hantavirus in the South-east coastal area of China

Objective To analyze the sequence of Hantavirus(HV) from Zhejiang, Fujian provinces and Shanghai municipal. Methods The sequence of M segment of several HV strains from coastal areas of Zhejiang, Fujian province and Shanghai municipal were collected and analyzed by Mega 4.0 and DNAStar software, phylogenetic trees were built by neighbor joining method (NJ) and analyzed to compare the similarity of HV strains isolated from Zhejiang, Fujian provinces and Shanghai municipal. Results The phylogenetic tree revealed that the similarity of isolates in the same province is much higher than the other, meanwhile, in same province or city, the nucleotide similarity of the same region is higher than the other regions. The distributions of the isolates from closer regions share higher sequence similarity in the phylogenetic branches. The ZH53 strain of HTN isolated from Fujian province, Gou3 strain and ZJ5 strain of SEO isolated from Jiande region in Zhejiang province formed a distinct phylogenic lineage in HTNV clade and SEOV clade respectively, and these strains were different from other variants and international standard strains with the similarity of 82.7%-86.3% and 84.0%-85.3% respectively. The SEOV strains in Zhejiang were isolated from field mouse indicating the phenomenon of “host spillover”. Conclusion The sequence similarity and the phylogeny of HV in southeast costal area of Zhejiang, Fujian provinces and Shanghai is closed related to the isolated regions, indicating geographic distribution of HV.

The function of histidine kinaseⅡ(Hk2) in the life cycle of Borrelia burgdorferi

Objective To validate the biological function of Hk2,which is reported to be in combination with the cognate response regulator Rrp2 to constitute a two component signal system in Borrelia burgdorferi that controls the key virulence factors required for pathogen invasion in mammals. Methods A hk2 knockout strain (hk2-) was generated through homologous recombination, with the biological function of Hk2 estimated by the mouse-tick animal model. Results The hk2- knockout strain remained to be able to infect mouse by needle inoculation, and subsequently successfully detected in the midgut of fed-larvae and -nymph by IFA. The quantitative-PCR demonstrated that hk2 knockout had no influence on bacterial replication in the midgut of fed nymph. However, only 50% of tested mice were infected in hk2- group by tick bite. Conclusion The hk2 knockout does not affect the pathogen acquisition from mouse by larvae ticks and subsequent persistent colonization in tick midgut, but it embarrasses the transmission from nymph to mouse under natural conditions by tick bite. The above data indicate that the Hk2 may effectively contribute to the pathogen invading mammals in the life cycle of B. burgdorferi.

Investigation of Bartonella infection in rats by molecular technology

【Abstract】 Objective To investigate the infection of Bartonella in rat?shape animals by molecular technology. Methods Rat?shape animals were captured by night trap in Tiantai county of Zhejiang province. DNA from rat livers were detected by PCR, and the positive products were sequenced. The infection rate of Bartonella in rat was calculated. Results There were 25 positive samples among 55 samples collected. The positive rate of Bartonella infection in Apodemus agrarius and Rattus losea were 48.84% and 33.33%, respectively. The sequence was more similar to B.doshiae. Conclusion The infection rate of Bartonella in rat?shape animals was higher in Tiantai county, and there was risk of transmitting this pathogen to human. The control measure should be figured out. It should take measures to control it.
